The start of ANEEL’s experimental development stage and TRL#4 are achieved with prototyping. In our case, defined as an adult genetically modified mussel. This translates into its feasibility, showing that silencing the gene we have chosen to modify, as well as inserting the CrISPR gene-drive construct, does not prevent or seriously disrupt any other vital function.

The challenges of this stage continue to be the result of the many gaps in our knowledge of this organism. The difficulty of replicating the reservoir environment in the laboratory (specifically the incredible abundance of food in eutrophied water) makes reproduction difficult. At the same time, some studies have shown that bivalves don’t have sex chromosomes (a hypothesis that we validated by constructing the mussel’s karyotype) and are not genetically determined, but can be affected, even reversed, by environmental factors such as temperature and food availability.

Without full mastery of reproduction, it is difficult to carry out experiments to determine haplossufficiency (the ability of a gene to determine a trait if it is only present in 1 copy – from the father or the mother) and thus unequivocally choose the gene for modification. We are trying to overcome this difficulty by studying reproduction and haplossufficient orthologous reproduction genes, as well as modeling the impact of environmental variables on populations.

This phase of the project also seeks to develop a methodology for assessing the severity of infestation in reservoirs, which is important for estimating the ratios of GM mussels to invaders needed for disinfestation in the future. This specific objective is being carried out by the Hubz company.
